How to Use ReceptoCraft

Getting Started

1. Create a free account at receptocraft.chordexbio.com. Use your .edu email for 30-day trial access.
2. Verify your email by clicking the link in the activation email.
3. Sign in and land on your Dashboard. Three tool cards are shown: Design, Create, and Develop.
4. To start a new session, click any tool card or use the "+ New" buttons in the header.
5. All sessions are saved manually — use the Save button in the top bar. You will be warned if you try to navigate away with unsaved changes.

Design Tool — Biologics Design

1. Select a construct type from the top tab strip: CAR-T, Fusion Protein, or Bispecific Antibody.
2. For CAR-T: Enter project name and description, then follow Steps 1–6. Step 1 requires VH and VL sequences in single-letter amino acid format.
3. Use the UniProt Lookup button (top right) to search for any protein by accession or name and retrieve its sequence and PTM features.
4. Click "Assemble Construct" in Step 5 to generate the full assembled sequence. This auto-populates the codon optimizer and PC calculator.
5. Step 5 tools include: Construct Assembler, Physico-chemical Calculator, Codon Optimizer, Sequence Translator, and RE Scanner.
6. Step 6 shows the Gene Strip, Protein Topology, and Color-coded Sequence Viewer (protein and DNA tabs).
7. Click Save in the top bar at any time to save your session to the cloud.

Create Tool — Gene Cloning

1. Enter your construct sequence (CDS, 5′→3′, DNA) in Step 1. The tool accepts IUPAC ambiguous bases.
2. Click "Analyse Sequence" to get GC content, ORF detection, feature annotation (if you specify domains), and restriction site scan.
3. Step 2: Select expression vector and system. The tool pre-fills cloning RE sites based on the vector MCS.
4. Step 3: Set primer design parameters (Tm, RE sites, Gibson overhang length) and click "Design All Primers".
5. Step 4: Assembly strategy is auto-selected based on insert size and fragment count. Override with the dropdown.
6. Step 5: Reagent list is auto-curated from your method and expression system. Add custom lab stocks.
7. Step 6: CrossRef literature search — terms are auto-generated from your construct type and insert name.
8. Step 7: Full sequence analysis including CAI, rare codons, liabilities, splice sites, and GC window chart.
9. Click Save to save the full session state to your account.

Develop Tool — R&D Project Manager

1. Click "+ New Project" to create a new R&D project file.
2. Each project has 7 tabs: Design, Materials, Protocols, Results, Process Flow, Reminders, and Report.
3. Design tab: Fill in hypothesis, objectives, acceptance criteria, assay strategy, and experimental design.
4. Materials tab: Log every reagent with vendor, catalog number, order status, QC, and storage details. The shipment tracker shows pending → ordered → shipped → received flow.
5. Protocols tab: Document each protocol step with SOP reference, operator, instruments, procedure, and QC result.
6. Results tab: Log each assay run with measurements, statistics, outcome vs. acceptance criteria, and interpretation.
7. Process Flow tab: Map the R&D pipeline with stages, milestones, assignees, and % complete.
8. Report tab: Auto-generated structured technical report. Add sign-off details (Prepared By, Reviewed By, Approved By).
9. Click "Save All to Cloud" to persist all projects. Projects auto-load from cloud when you open the Develop tool.

Account & License Management

1. Trial accounts (7 or 30 days) have full access during the trial period.
2. After trial expiry, purchase a license key from the ChordexBio store and activate it at receptocraft.chordexbio.com/upgrade.php.
3. One license key = one account. Keys cannot be shared.
4. To change your password or view account details, use Account Settings from the user menu (top right).
5. Contact support@chordexbio.com for billing issues, lost license keys, or technical support.
